The mango sudden death syndrome has become a serious threat to the mango industry and caused significant decline in mango production worldwide. The bark beetle Hypocryphalus mangiferae (Stebbing) (Coleoptera: Curculionidae: Scolytinae) has been suggested as a potential vector of the disease based primarily on field observations with little or no supporting empirical data. In this study, we investigated the role of infected mango trees in host attraction and colonization by H. mangiferae to determine if beetle attack and colonization contributes to the disease progression on mango trees. Initially, the role of various stress factors on beetle attraction and disease progression was assessed under lathe house conditions from 2008 to 2009. Results suggest that symptomatic or recently inoculated mango trees (without any obvious symptoms) are preferentially colonized by H. mangiferae. Although not significant, high numbers of beetles attacked stressed or wounded mango trees, compared to healthy or dead mango trees. Disease symptoms after beetle colonization, such as bark splitting, wilting and oozing, were further evaluated. These symptoms showed positive correlation with the degree of disease severity and host plant condition. Furthermore, two fungi, Ceratocystis fimbriata and Lasiodiplodia theobromae, were frequently isolated from the beetle and beetle-colonized trees. Based on these findings, they suggests that H. mangiferae can vector multiple fungi associated with mango sudden decline disease and play a significant role in outbreaks of this disease.  相似文献

The analysis of structural variants associated with specific phenotypic features is promising for the elucidation of the function of involved genes. There is, however, at present no approach allowing the rapid mapping of chromosomal translocation breakpoints to the basepair level from a single chromosome. Here we demonstrate that we have advanced both the microdissection and the subsequent unbiased amplification to an extent that breakpoint mapping to the basepair level has become possible. As a case in point we analysed the two breakpoints of a t(7;13) translocation observed in a patient with split hand/foot malformation (SHFM1). The amplification products of the der(7) and of the der(13) were hybridized to custom‐made arrays, enabling us to define primers at flanking breakpoint regions and thus to fine‐map the breakpoints to the basepair level. Consequently, our results will also contribute to a further delineation of causative mechanisms underlying SHFM1 which are currently unknown.  相似文献

Many phocids are capital breeders, relying on stored reserves to sustain energetic requirements while on land. Their large body size, high energy expenditure during lactation, and the insulative effects of the blubber layer can lead to thermal stress from overheating, especially in warm and temperate climates. Thermal stress can influence fine‐scale site choice on breeding colonies, and behavioral thermoregulation has been proposed as an explanation for the clear preferences shown by breeding female gray seals for proximity to pools of water. However, anecdotal observations suggest that pools of water may also be preferred for drinking, though water intake is difficult to verify without real‐time physiological monitoring. Here, an alternative approach demonstrates that gray seals also require access to water for drinking. Using Ecological Niche Factor Analysis to examine fine‐scale physical determinants of pupping site choice at North Rona, Scotland, we found that lactating mothers showed preference for lower salinity pools. This is most pronounced early in the season, when ambient temperatures and presumably thermal stress are greatest. Given that the cooling effect of fresh and salt water should be equivalent, the most parsimonious explanation for this preference for fresh water pools is that lactating females use these pools for drinking.  相似文献

1 Larval and adult Hylobius abietis (L.) can feed and, in the case of the adults, cause commercially significant damage to many species of conifer. The present study aimed to evaluate the impact of larval and adult feeding on different host plants on subsequent reproductive fitness. 2 Hylobius abietis larvae were reared on logs of four host species known to vary in suitability for development; Pinus nigra ssp. laricio, Picea sitchensis, Larix kaempferi and Pseudotsuga menziesii. Adult females were collected on emergence and allocated to complete maturation feeding on one of four seedling conifer species, P. nigra spp. laricio, P. sitchensis, P. menziesii and Larix ×marschlinsii. It was assumed that variation in both larval and adult host plant quality had the potential to influence subsequent reproductive behaviour. 3 The time taken for females to attain reproductive maturity was assessed, and reproductive output was measured over an average period of 17 days. 4 The best predictor of the time to attain reproductive maturity was found to be the initial adult body weight. This was strongly related to the larval host species, with the largest adults emerging from P. nigra ssp. laricio. The time taken to begin oviposition was in the range 10–49 days (mean 23 days). The subsequent mean rate of oviposition for individual females was in the range 0.1–3.7 eggs per day. Egg volumes varied between and within individual females, in the range 0.241–1.079 mm3. 5 The species of seedling provided for maturation feeding had no significant impact on reproductive behaviour over the course of the experiment. It is possible, however, that the quality of the adult diet might exert a greater influence on behaviour over a longer period because H. abietis is a relatively long‐lived species.  相似文献

Bone mineralization is strongly stimulated by weight-bearing exercise during growth and development. Judo, an Olympic combat sport, is a well-known form of strenuous and weight-bearing physical activity. Therefore, the primary goal of this study was to determine the effects of Judo practice on the bone health of male high school students in Korea. The secondary goal of this study was to measure and compare the bone mineral density (BMD) of the hands of Judo players and sedentary control subjects. Thirty Judo players (JDP) and 30 sedentary high school boys (CON) voluntarily participated in the present study, and all of the sedentary control subjects were individually matched to the Judo players by body weight. BMD was determined by using dual-energy X-ray absorptiometry (Hologic, Bedford, MA, USA). The lumbar spine, femur and forearm BMD in the JDP group were significantly greater by 22.7%, 24.5%, and 18.3%, respectively, than those in the CON group. In addition, a significant difference in the CON group was observed between the dominant hand (DH) radius (0.710 ± 0.074 g/cm2) and the non-dominant hand (NDH) radius (0.683 ± 0.072 g/cm2), but this was not observed in the JDP group (DH = 0.819 ± 0.055 g/cm2; NDH = 810 ± 0.066 g/cm2) (P < 0.05). Therefore, the results of this study suggest that Judo practice during the growth period significantly improves bone health in high school male students. In addition, it seems that Judo practice could eliminate the effect of increased BMD in the dominant hand.  相似文献

Feeding success depends on host availability, host defensive reactions and host preferences. Host choice is a critical determinant of the intensity at which pathogens are transmitted. The aim of the current study was to describe host preferences of Palaearctic Culicoides species (Diptera: Ceratopogonidae) Latreille using traps baited with the five different host species of poultry, horse, cattle, sheep and goat. Collections were carried out nightly in July and August 2009 in western France with three replicates of a 5 × 5 randomized Latin square (five sites, five hosts). Moreover, an ultraviolet (UV) light/suction trap was operated during host‐baited collections to correlate Culicoides biting rates and UV light/suction trap catches. A total of 660 Culicoides belonging to 12 species, but comprised mainly of Culicoides scoticus Downes and Kettle, Culicoides dewulfi Goetghebuer and Culicoides obsoletus Meigen, were collected on animal baits. Abundance was highest for the horse, which accounted for 95% of all Culicoides caught, representing 10 species. The horse, the largest bait, was the most attractive host, even when abundance data were corrected by weight, body surface or Kleiber's scaling factor. Culicoides obsoletus was the only dominant species attracted by birds. Both C. scoticus and C. dewulfi were collected mainly from the upper body of the horse. Finally, the quantification of host preferences allows for discussion of implications for the transmission of Culicoides‐borne pathogens such as bluetongue virus.  相似文献

Worldwide, farm animals are kept on litter or foraging substrate that becomes increasingly soiled throughout the production cycle. For animals like laying hens, this means that it is likely they would scratch, forage and consume portions of excreta found in the litter or foraging substrate. However, no study has investigated the relative preference of laying hens for foraging and consumption of feed mixed with different percentages of excreta. A total of 48 White Leghorn laying hens of two strains, a commercial strain (Lohmann LSL-Lite (LSL), n=24) and UCD-003 strain (susceptible to liver damage, n=24), were individually housed and given access to feed mixed with increasing percentages of hen excreta (0%, 33%, 66% and 100% excreta diets) and corn as a luxury food reward (four corn kernels per diet daily). The amount of substrate and number of corn kernels consumed from each diet was recorded for a period of 3 weeks. Both LSL and UCD-003 hens preferred to consume and forage in diets with 0% excreta, followed by 33% and finally diets containing 66% and 100% excreta. Despite the presence of excreta-free diets, birds consumed on average 61.3 g per day of the diets containing excreta. Neither physical health, measured by plasma enzyme activity levels, nor cognitive differences, assessed by recalling a visual discrimination task, was associated with relative feeding or foraging preference. In conclusion, this study demonstrated a clear preference for feeding and foraging on substrate without excreta in laying hens. However, considering the amount of excreta diets consumed, further studies are needed to understand the causes and consequences of excreta consumption on physiological and psychological functioning, and how this information can be used to allow adjustments in the management of foraging substrates in farmed birds.  相似文献
